Sample Collection Procedures

Let's begin with water sampling materials. Protocols for collecting and storing samples can be next and then we can move onto soil.


In-line water sampling materials, starting from the target water source :


One length of 1/4 inch sterile tubing.

One Smith-Root .45 micron single-use filter packet.

One length of sterile tubing.

One handheld non-sterile vacuum pump capable of creating at least 10hg of negative pressure.

One length of non-sterile tubing.

One rubber stopper with two through holes. One for discharge water entering the bottle, and one to let air escape.

One clear plastic one liter bottle, or a larger clear plastic container marked in one liter increments for measuring how much water has been pumped.


Note: If one is going to collect water into a container, bring it to a stream bank and THEN run it through the filtering system, the container must be a sterile Nalgene container. 


Materials for transporting and setting up system components on-site.


One container with a lid, sterile on the inside, for storing sterile tubing and filter pack.

One or two pairs of non-powdered nitrile gloves for handling sterile components during assembly and deployment into a water source.


So basically, all components from the water source to the pump need to be sterile. Nothing after that, including the pump, needs to be sterile. 

A way to support the one liter water discharge bottle so that it remains upright even when pulled on by the tubing would be a good thing to do.

Collecting a dry sample is a much easier process.        The most stringent protocol would be to wear a mask (we have to wear one in WA anyway), wear sterile gloves,  sterilize the tweezers with a flame,   insert the sample into a sterile glass or paper DNA sample container.
